C natural abundance: rooting medium matters

چکیده
• Tree root respiration utilizes a major portion of the primary production in forests and is an important process in the global carbon cycle. Because of the lack of ecologically relevant methods, tree root respiration in situ is much less studied compared with above-ground processes such as photosynthesis and leaf respiration. • This study introduces a new 13 C natural tracer method for measuring tree root respiration in situ . The method partitions tree root respiration from soil respiration in buried root chambers. • Rooting media substantially influenced root respiration rates. Measured in three media, the fine root respiration rates of longleaf pine were 0.78, 0.27 and 0.18 mg CO 2 carbon mg − 1 root nitrogen d − 1 at 25 ° C in the native soil, tallgrass prairie soil, and sand–vermiculite mixture, respectively. Compared with the root excision method, the root respiration rate of longleaf pine measured by the field chamber method was 18% higher when using the native soil as rooting medium, was similar in the prairie soil, but was 42% lower if in the sand–vermiculite medium. • This natural tracer method allows the use of an appropriate rooting medium and is capable of measuring root respiration nondestructively in natural forest conditions.

Comparative performance of in vitro multiplication in four grape (Vitis spp.) rootstock genotypes
The magnitude of demand for planting materials in grape, mainly for rootstock genotypes indicates that micropropagation is inevitably necessary for their mass scale propagation. Therefore, the studies on micropropagation of four genetically different grape rootstocks namely Dogridge (Vitis champini), SO4 (V. riparia× V. berlandieri), H-144 (V. vinifera × V. labrusca) and 3309 C (V. riparia × V....
متن کاملRequirements for in vitro rooting of Quercus robur and Q. rubra shoots derived from mature trees.
Stabilized shoot cultures initiated from crown material of six adult Quercus robur L. trees and from basal epicormic shoots of a Quercus rubra L. tree showed good in vitro rooting capacity. An initial five-day dark period generally improved the rooting response but was detrimental to plantlet quality. There were clonal differences in rooting capacity. The concentration and exposure time of the ...
متن کاملEffects of Liquid Medium on Rooting and Acclimation of Regenerated Microshoots of Banana (Musa sapientum L.) cv. Sagar
A regeneration protocol ensuring a high frequency rooting of micro-shoots derived from apical and axillary buds of suckers of banana cv. Sagar was achieved by using liquid medium. When the explants were cultured on MS medium supplemented with 0.5 mg/l each of BA, Kn and NAA, a large number of shoots developed. With the progression of the number of subcultures, shoot proliferation was enhanced. ...
متن کاملAn efficient regeneration and rapid micropropagation protocol for Almond using dormant axillary buds as explants.
An efficient in vitro protocol was standardized for Almond (Prunus dulcis) propagation using dormant axillary buds as explants. Explants were cultured on Murashige and Skoog (MS) and woody plant medium (WPM) supplemented with different concentration/combination(s) of phytohormones. MS basal medium showed lowest shoot induction and took longest duration for shoot initiation. Multiple shoots were...
